You seem to be very clear on how the GUI should look and what features it should contain. This is a great advantage. Regarding GPS over RS232, there are standard PyPI modules to help. I expect the bulk of the work will involve the GUI and not the GPS interface. For the GPS interface I would propose writing an adapter. By implementing an adapter design pattern you can define "virtual" adapters that feed test data. Your project description lacks detail, so I am not certain if I would have to setup a Raspberry PI with a GPS over RS232 receiver for development purposes. I am willing to do this. Note that unless you have a specification already written for the software, I would have to start with writing a spec. Your requirements are not trivial, and starting without a clear specification will cause serious delays later and my lead to a product that does not deliver exactly what you wanted.
